(a) Draw and briefly explain three different release profiles of different modified release dosage forms (the figures should be correctly labelled showing axes scales and units).

(b) Critically assess a formulation that you would use to achieve one of the release profiles that you mentioned in previous part of this question. Draw the corresponding pharmacokinetics profile of this release pattern and mention its clinical application(s).

(a) Release profiles of modified release dosage forms:
Immediate Release: This profile is characterized by rapid release of the drug, usually within 30 minutes to an hour. The graph would show a sharp increase in drug concentration, followed by a steady decrease as the drug is metabolized and eliminated. Graph: Y-axis (Drug concentration), X-axis (Time) - Rapid increase in drug concentration, followed by a steady decrease.
Sustained Release (SR) or Controlled Release (CR): The drug is gradually released over an extended period of time. This is designed to maintain a constant drug concentration in the bloodstream, avoiding peaks and troughs associated with immediate release forms. Graph: Y-axis (Drug concentration), X-axis (Time) - Gradual increase in drug concentration, followed by a plateau and then a slow decrease.
Delayed Release: The drug release is postponed until it has passed through the stomach to avoid the acidic environment, or for targeted release in specific areas of the gastrointestinal tract. The graph would show a lag phase with no drug release, followed by a rapid increase as the drug is released. Graph: Y-axis (Drug concentration), X-axis (Time) - No drug release initially (lag phase), then a sharp increase, followed by a decrease.
